No. Which frame, which area do you want to drill ??
Never. I won’t compromise the strength of the frame. An external cable routings may be necessary sometimes.

Never drill a hole in a frame, you could potentially widen one but any ports/holes on a frame are structurally accounted for in the initial build.
The double triangle bike design is still the strongest structure since the 1800's, it has not changed.
